Constitutive knockout mice are engineered to carry a gene that doesn’t work. Typically, the inactivation of a gene is achieved by deleting (part or all) of its sequence, and it is nonfunctional in the whole animal. Conditional knockout mice provided by Creative Biogene allow gene deletion in a tissue- and/or time-specific manner. Typically, conditional KO mice are achieved through the Cre-lox system. Instead of deleting key sequences, it is flanked by loxP sites (called floxed sequences). Cre recombinase deletes the sequence between the two loxP sites. Inducible or tissue-specific Cre is used to knock out gene function in that tissue only.

Conditional knockout/inducible mice, inducible knockout mice and knockdown mice from Creative Biogene avoid the risk of lethal phenotypes associated with gene inactivation at the early stages of development and allows analysis of the effects of gene inactivation in adult animals. If an unexpected phenotype emerges in a knockout mouse population, Creative Biogene researchers using appropriate wild-type and/or HE controls will help assess where exactly the phenotypic changes in knockout mice come from. If the phenotype is induced by a mutated target gene, the control should not show that phenotype.

Creative Biogene creates new knockout mice through CRISPR/Cas9* genetic engineering, always backcrossing the first KO mouse female with a wild-type male to refresh/swap the Y chromosome. Then in subsequent breedings, the transgenic males obtained from the first round of breeding are used to mate with wild-type females.
